Sample Submission

Prior to sending any samples to the lab, please ensure that a GIGPAD batch has been created (http://portal.hpcgg.org) and please include the batch paperwork with any samples.

Sanger sequencing is performed using the 96-capillary ABI 3730xl DNA Analyzer. This is the "Gold Standard" for producing superior data for high throughput sequencing and genetic analysis. The machine is highly sensitive and reliable. It also has a stable automated system that can operate unattended for up to 48 hours, thus minimizing errors.
This facility is equipped with two ABI 3730xl DNA Analyzers. They provide reads of up to 800 high quality bases and produce approximately 1500 reads per day, per machine (1.2 Megabases of sequence per machine).

 

 

Submission Requirements

We are only able to accept customer submission of ready-to-run plates for sequence detection.  The products can either be cleaned and lyophililzed and submitted in 96-well plates for the Core to resuspend in Hi-Di formamide, or 96-well or 384-well plates of cleaned sequencing reaction product resuspended in water or formamide ready to run for detection.

Suitable 96-well plates:

Thermo-Fast 96 Detection Plate (Fisher Scientific, Cat# AB-1100).

Please seal all plates using adhesive PCR film (Abgene AB-0558).

 

Suitable 384-well plates:

Thermo-Fast Diamond 384 (Fisher Scientific, Cat # AB-1111).

Please seal all plates using adhesive PCR film (Abgene AB-0558).

Please also ensure wells are left empty in either plate submission so we can add in our controls.
If you submit a 96 well plate, you MUST keep well A1 empty.
If you submit a 384-well plate, you MUST keep the following wells empty:

 

 

 

 

Quadrant 1 =   A01, O2

Quadrant 2 = A02, O24

Quadrant 3 =   B01, P2

Quadrant 4 = B02, P24

 

All plates must be clearly labeled with sample names as entered on the relevant sample submission form. Print the form and submit along with samples and also email the form to heyi@partners.org. An electronic copy MUST be emailed.

Pack plates in dry ice in a styrofoam (or other insulated) cooler. Make sure there is dry ice on the top, bottom, and both sides of the plates. If shipping more than 1 plate, wrap plates in aluminum foil and/or vinyl tape so they will not move around during shipment.

Please be aware that sample names in the Sample Submission form that is uploaded in GIGPAD must contain only alphanumerics (i.e., no underscores, dots, dashes, spaces, etc.). This ensures that the detection and any downstream analysis pipeline processes run smoothly.

Please note we are a high throughput sequencing facility, so we do not accept single sample submissions. Our sequencers detect 96 reactions (1 quadrant) in parallel and up to 384 reactions (4 quadrants) per plate. If you submit an order for less than one quadrant, please note you will still be charged the price of one quadrant for sequencing.

For purposes of planning sequencing runs, we require that customers notify us a day in advance when they plan on submitting plates, and for the plates to submitted no later than 10 AM on the day if they are to be run on that day.
